Alexandra Daddario spielte von 2002 bis 2003 in 50 Folgen der "Seifenoper All My Children" die schlecht behandelte Laurie Lewis und beendete gleichzeitig ihre Schulausbildung an der PCS (Professional Childrens School). Anschließend trat sie abwechselnd im Fernsehen und auf der Leinwand auf. 2005 übernahm sie in dem Film "Der Tintenfisch und der Wal" (The Squid and the Whale) eine Nebenrolle. Im Jahr 2009 war sie in dem Film Jonas Brothers: Das ultimative 3D Konzerterlebnis zu sehen.Daddario absolvierte auch einige Gastauftritte, so in "Die Sopranos" (The Sopranos), "Criminal Intent-Verbrechen im Visier" (Law and Order: Criminal Intent),"Law & Order", "Conviction" und"Life on Mars" (Life on Mars).2010 spielte sie in dem Fantasyfilm"Percy Jackson-Diebe im Olymp" die Rolle der Annabeth Chase.
